Hinokitiol (p-thujaplicin) to substancja wystepujaca naturalnie w zdrewnialych czesciach niektorych drzew - hinokito, czyli cedru japonskiego, europejskiego czerwonego cedru i kilku innych. jest silnym naturalnym chelatorem jonow metali, ponadto trzepie niechciane mikroby, w Japonii to oficjalnie zarejestrowany bezpieczny dodatek do zywnosci.

In this study, we examined the modulatory effect of hinokitiol (HK) on the production of tumor necrosis factor (TNF)-alpha, a critical factor involved in skin inflammation and hair follicle apoptosis. HK effectively suppressed TNF-alpha production in lipopolysaccharide (LPS)-activated, macrophage-like (RAW264.7) cells. This compound also diminished mRNA synthesis of TNF-alpha, indicating that HK-mediated inhibition may occur at the transcriptional level. Moreover, this compound down-regulated the phosphorylation of PDK1, Akt/PKB, and ERK, resulting in a loss of nuclear factor (NF)-kappaB activation, which is detectable by immunoblotting and reporter gene assays. Therefore, these results suggest that HK may cure hair loss by suppressing factors that promote follicular apoptosis, such as TNF-alpha, in addition to stimulating new hair growth.

Nanocapsules containing hinokitiol (HKL) were prepared by an emulsion-diffusion method. In an emulsification step in preparing nanocapsules, cetyltrimethylamonium chloride (CTAC) was employed as a cationic emulsifier, Poly(epsilon-caprolactone) (PCL) was use as a wall material and HKL dissolved in octylsalicylate (OS) was used as a core material. The submicron-sized nanoparticle was observed on a TEM. The size ranged 55-234 nm and the mean diameters were 223 nm, which were determined by a dynamic light scattering method. According to the results of pH-dependent microelectrophoresis, the absolute value of the surface potential of the nanocapsules was greater than 20 mV. The nanocapsules were colloidally stable over the pH range of 3-11. The nanocapsules were included in two kinds of preparations, namely shampoo and hair tonic, and the preparations were applied every day for 3 weeks on the clipped backs of 6 week-old mouse (C57BL/6) to investigate the hair growth-promoting effect. The degree of hair growth was evaluated by image-analysing the photographs of the backs and, in parallel, by the histological observation of the formation and the growth of hair or hair bulbs. The results were compared with those of commercially available Minoxidil solution (3%). Phosphate buffered saline was used as a control. The in vivo hair growth-promoting effects of the two preparations were comparable to those of Minoxidil solution. These results are in a good agreement with the histological and structural changes of follicles of the model animals, of which skins were treated with either the testing samples or the control in the same way the experiments of in vivo hair growth promotion were performed.

Hinokitiol (beta-thujaplicin), a troplone-related compound found in the heartwood of cupressaceous plants, strongly inhibits the proliferation of a broad range of tumor cell lines. This is the first report to demonstrate that hinokitiol, a metal chelator derived from natural plants, suppresses cell growth and disrupts AR signaling in prostate carcinoma cell lines. Our present studies indicate that hinokitiol suppresses androgen/AR-mediated cell growth and androgen-stimulated DNA synthesis by [(3)H]thymidine incorporation in a dose- and time-dependent manner. Hinokitiol simultaneously suppresses the intracellular and secreted PSA levels, a marker for the progression of prostate cancer. Hinokitiol significantly represses the AR mRNA and protein expression in a dose- and time-dependent manner. Additionally, the ligand-binding assay shows that hinokitiol blocks binding of the synthetic androgen [(3)H]R1881 to AR in LNCaP cells. These findings collectively suggest that hinokitiol is potentially effective against prostate cancer in vitro, and thus it might become a novel chemopreventive or chemotherapeutic agent for prostate cancer.

In vitro permeation of hinokitiol (HKL) through hairless mouse skin was investigated using a diffusion cell. Either propylene glycol (PG) or ethanol (EtOH) was used as a vehicle for HKL. After applying the HKL solutions of 0.5%. 1%, 2%, and 5% onto the skin, the amount of HKL transferred through the skin into the receptor solution, phosphate-buffered saline (PBS, pH7.4), was determined at a predetermined time intervals for 18 hr using a high performance chromatography (HPLC). EtOH was more effective than PG in terms of in vitro permeation of HKL. This is possibly because EtOH acts as a permeation enhancer. Another reason would be related to the higher thermodynamic activity of HKL in ethanol. To investigate the effect of an enhancer on the in vitro permeation, oleyl alcohol, 1-dodecyl-2-pyrrolidone (DP), and lauric acid were used as enhancers. Each was added to the HKL solution (1%) so that the concentration of the enhancer was 1%. Among the enhancers, DP was the most effective and it enhanced the permeation of HKL approximately 5-10 times.
